ABSTRACT: Ovarian cancer is a malignant cancer cause of death for most women in Indonesia. The emergence of ovarian cancer showed no specific symptoms in its early stages, so that ovarian cancer screening mostly when patients have an advanced stage. Treatment of ovarian cancer at an advanced stage will be more difficult with a poor prognosis. Minimally invasive biomarkers that are needed to be able to diagnose cancer at an early stage. One biomarker is secreted in the blood vessels that microRNA The expression patterns between microRNA and mRNA can be used as clinical information on ovarian cancer. One microRNA whose existence is linked to the incidence of cancer is hsa-miR-21-5p. The role of hsa-miR-21-5p as onkomir namely silence or degrade the target mRNA. Based on the analysis insilico, RECK which acts as a tumor suppressor is the target of a hsa-miR-21-5p. This study aims to determine the expression of hsa-miR-21-5p in plasma samples EOC patients compared with healthy controls. Blood samples obtained plasma was isolated, and isolation of RNA from plasma using miRCURY RNA Isolation Kit - biofluids. RNA obtained is then synthesized cDNA using miRCURY LNA microRNA cDNA synthesis kit II. cDNA were then quantified using real-time qPCR machine (Biorad CFX 96) by reacting Exilent SYBR Green master mix, primer microRNA and cDNA. Quantification of mRNA using a kit KAPA SYBR FAST q Universal One-Step RT-PCR kit. Results show hsa-miR-21-5p in patients with EOC increased expression as much as 4.7579-fold compared with healthy controls and statistical analysis showed a significant result (p <0.05), whereas the RECK expression decreased 4,2 times with foldchange 0.237665 on plasma EOC patients compared with healthy controls and statistical analysis showed a significant result (p <0.05). The result of using the Spearman rho correlation analysis shows that the expression hsa-miR-21-5p relationships inversely proportional to mRNA Reck with strong correlation. This study has been able to prove that the HSA-miR-21-5p undergo overexpression in EOC patients were accompanied by downregulation of RECK expression than in healthy controls. Hsa-miR-21-5p also expressed in the plasma, so as to develop a minimally invasive method for the diagnosis of cancer incidence.
