MDM2 polymorphism SNP309 T> G; Study of Early Onset Breast Cancer Risk

ABSTRACT: Introduction: The incidence of early onset breast cancer is increasing in Indonesia. Early onset breast cancer usually has poorer prognosis and high level of recurrence. MDM2 SNP309 T>G as a risk for early onset breast cancer has been observed in several studies with contradicting results among different races. Study related to MDM2 SNP309 T>G polymorphisms in early onset breast cancer has not yet done in Indonesia. Methods: DNA from blood samples were obtained from 104 early onset breast cancer patients (<40 years old) and 102 breast cancer patients older than 55 years old registered at RS Dr. Sardjito in the period of 2006-2012. MDM2 SNP309 T>G polymorphisms were detected using PCR-RFLP method. Chi-square was used to analyze the difference in proportion of genotype and allele with the associate risk to early onset breast cancer. Result: Proportion of genotypes among breast cancer patients in Yogyakarta were 20,9%, 52,4%, and 26,7% for T/T, T/G and G/G, respectively. T/G was higher in early onset (29,1%) than late onset (23,3%), but was statistically not significant(p>0,05). Allele T was found significantly higher in early onset patients (p=0,0436). Conclusion: There is no relationship between MDM2 SNP309 T>G polymorphism and early onset breast cancer in Yogyakarta.
