INFECTION Human papillomavirus (HPV) 16/18 AND MOVEMENT p53 exon 7 EVENTS IN HEAD AND NECK CANCER

ABSTRACT: Indonesia National Agency for Cancer Registration, puts head and neck cancer on fourth from ten malignancies in men and women. Head and neck cancer caused by several factors, like alcohol consumption, smoking, virus infection, diet and genetic mutation. Human papillomavirus 16/18 infection found in several head and neck cancer cases. Generally, head and neck cancer with HPV positive does not associated with p53 mutation. In the other hand, head and neck cancer with HPV negative ussually associated with p53 mutation. Most of p53 mutation have been reported in p53 exon 5-8 and the most frequently mutated in exon 7. Purpose of research are examine the associate between HPV 16/18 infection in head and neck cancer and p53 exon 7 mutation using Polymerase Chain Reaction and sequencing. Reserach was done in Dharmais Cancer Hospital Jakarta. There are 53 samples from 37 patients, consist of 33 Formalin Fixed Paraffin Embeded (FFPE) samples, 14 fresh tissue samples and 6 blood samples. Result of HPV typing indicated 12 FFPE samples and 2 fresh tissue samples that HPV 18 positive. Analysis result of sequencing of p53 exon 7 from 8 FFPE samples, 5 fresh tissue samples and 5 blood samples showed no mutation were detected. HPV 18 is assosiated with head and neck cancer. Several problem which can not analysed from this researches are possibility mutation of p53 occur in besides exon 7 or sampel infected by another serotype of HPV.
