ANTICANCER ACTIVITIES OF ALFA terpineol SAP Pinus (Jungh. Et de Vriese) ON CELL CYCLE, APOPTOSIS AND EXPRESSION OF PROTEIN (Bcl-2 and Bax) CELL LINE MCF-7

ABSTRACT: Background: Previous study showed that alfa terpineol from Pinus merkusii had cytotoxic effet on several cancer cell line namely T47D and HeLa. Objective: The aim of this study was to determine the effect of alfa terpineol on cytotoxicity, cell cycle changes, apoptotic and the changes expression of Bcl-2 and Bax in MCF-7 cell Methods: The cytotoxicity activity of alfa terpineol was determined using MTT assay, where the IC50 value was used as a parameter for cytotoxicity. Cell cycle changes was detected by Pi staining and flowcytometri. Annexin V-Pi staining and flowcytometry was used for detecting early apoptotic cell. The expression of Bcl-2 and Bax protein was explored by immunocytochemistry. Result: MTT assay showed that alfa terpineol inhibited the growth of MCF-7 with the IC50 value of 33μg/ml. Treatment using the dosage of the IC50 of alfa terpineol increased cell accumulation in Sub-G1 phase and caused early apoptotic cell 5,92% compare to control. Evaluation of alfa terpineol decreased level of Bcl-2 and increased level of Bax. This result showed that alfa terpineol induce apoptosis through mechanism via mitochondrial pathway. Conclusion: It is showed that alfa terpineol has the potential as anticancer agent and its mechanism are through inhibition of cell cycle and stimulation of apoptosis.
