INCREASING THE SENSITIVITY OF CANCER CELLS KOLON WiDr AGENT 5-fluorouracil chemotherapy ethanolic GRASS BY EXTRACT PEARL (Hedyotis corymbosa L.)

ABSTRACT: Pearl grass (Hedyotis corymbosa L.) is a wild grass that can be found anywhere therefore have a high economics value when it's used as cochemotherapeutic agent. Combination of Hedyotis corymbosa ethanolic extract (HEE) with doxorubicin showed apoptosis induction on MCF-7 breast cancer. On colon cancer which have p53 mutant causing this cancer resistant to 5-fkuorouracil (5-FU) and decreased the 5-FU efficiency. The aim of this study is to know the cytotoxic effect and apoptotic induction of HEE and 5-FU combination on WiDr colon cancer. HEE got from maceration by using 96% absolute ethanol. Active compound was analyzed by using thin layer chromatography (TLC). Cytotoxicity assay was conducted by using MTT assay for both single and combination treatments for 24 and 48 hours. IC50 and CI are parameters for cytotoxicity and synergysitic combination. Apoptotis induction was analyzed by using flowcytometry Annexin V-PI. TLC profile showed that HEE contain ursolic acid (hRf 60). Treatment of HEE and 5-FU on WiDr cells for 24h haven't showed optimum cytotoxic effect (IC50 450 Ã���Ã��Ã�¼g/ml and undetermine), while the treatment for 48h showed cytotoxic activity with IC50 80 Ã���Ã��Ã�¼g/ml and 20 Ã���Ã��Ã�¼M. Combination of HEE and 5-FU for both 24 and 48 h showed synergistic effect with CI<0.9 (n=3; p<0.05). Combination of HEE (20 Ã���Ã��Ã�¼g/ml) and 5-FU (5 Ã���Ã��Ã�¼M) for 24h increased the cell death through apoptosis and necrosis compared to their single treatment.
