The number of colon cancer case increased significantly in the recent years. Decreasing cancer risk could do with prevention. One of the candidates that can be used for cancer preventive food is black rice that contains several bioactive coumpounds especially antosianin. The purpposes of the riset were to determine the cytotoxic activity of ethanolic extract and fractions of black rice bran Wojalaka that has limited information, to identify the groups of coumpounds which have cytotoxic activity, to know inhibition of proliferation, and also to find out the death process cell and cell cycle profiles of colon cancer WiDr. The research was began with the extraction of rice bran Wojalaka by maceration using ethanol followed by Thin Layer Chromatography (TLC) to monitor the coumpound content. After that, fractionation processed by Preparative Thin Layer Chromatography (PTLC). The each fraction was used for cell culture cytotoxicity assay WiDr and Vero cells using MTT Assay. The most toxic fraction was used to identify the bioactive class coumpound, seen the antiprolifetive effect and observed cell death and cell cycle profile WiDr Using Flowcytometri. IC50 values were quantified using probit analysis. Results showed that the ethanolic extract had cytotoxic activity with IC50 620.66± 61.57 µg/ml. From 6 fraction, F6 was the most toxic fraction with IC50 44.01± 0.86 µg/ml. The coumpund groups which have cytotoxic activity wetr phenolic, flavonoid, terpenoid, steroid and was estimate had kuersetin, F6 was able to inhibited proliferation cell, induce apoptosis further and caused G2/M-phase arrest on WiDr cell line.
