Combination Effects search ethanolic extract of leaves Jure (Nerium indicum Mill.) And distillate Cinnamon (Cinnamomum burmanii) in Cytotoxic Test and Test of Breast Cancer Cell Migration
ABSTRACT: Metastasis is the major problem in cancer patient's death. One of the stages of metastasis is the cancer cells migration. Jure (Nerium indicum Mill.) and cinnamon (Cinnamomum burmannii) have anticancer activity. The aims of this study was to determine the cytotoxic effects and inhibition of breast cancer cell migration by combination of jure leaf ethanolic extract (JLEE) and cinnamon essential oil (CEO). JLEE compounds was determined by using thin layer chromatography (TLC), while CEO compound was determined by using gas-chromatography mass spectrometry (GC-MS). Cytotoxic effect was observed by using MTT assay, while JLEE-CEO combination effect on cell migration was observed through scratch wound healing assay. TLC and GC-MS profile showed that JLEE contains cardenolida and CEO contains sinamaldehid. This study showed that JLEE and CEO are toxic in 4T1 cells with IC50 values are 124 mg / mL and 2,4 mg / mL respectively. Combination of JLEE and CEO showed a synergistic effect and decrease the 4T1 cell viability to 69%. Further analysis on the cell migration showed that JLEE and CEO tended to inhibit cancer cells migration. The result conclude that JLEE and CEO have cytotoxic effect and tends to inhibit cancer cells migration.
