SEARCH THE POTENTIAL cytotoxic ethanolic GRASS EXTRACT PEARL (Hedyotis corymbosa L.) AGAINST BREAST CANCER CELLS HER2 protein overexpression

ABSTRACT: Breast cancer is one of cancer types with high incidence in the world, and 20% of the case are caused by HER2 (Human Epidermal Growth Factor Receptor 2) overexpression. Inhibition of HER2 overexpression becomes a promising strategy in breast cancer therapy. Hedyotis corymbosa L., who contains ursolic acid which is known for its activity in inhibition of NF-Ã��Ã�ºB as a part of HER2 downstream, thus it is predicted that Hedyotis corymbosa L. possesses cytotoxic effect through HER2 inhibition. This study aims to investigate cytotoxic activity of Hedyotis corymbosa Ethanolic Extract on HER2 overexpression breast cancer cells (MCF-7/HER2). Hedyotis corymbosa Ethanolic Extract (HEE) was made by maceration with 96% of ethanol. Ursolic acid in HEE was identified qualitatively by using Thin Layer Chromatography (TLC). The cytotoxic activity of HEE against MCF-7/HER2 was carried out by MTT assay with IC50 value as parameter. The activity of HEE in HER2 expression on MCF-7/HER2 cells was observed by immunofluorescence assay and analyzed as the lower fluorescent intensity means the lower expression of HER2. According to TLC, it was indicated that HEE had similar hRf score with ursolic acid standard as 60. From cytotoxic assay, HEE had cytotoxic activity towards MCF-7/HER2 in dose dependent manner with IC50 value of 696 Ã��Ã�µg/mL. Furthermore, HEE showed the lower expression of HER2 expression as compared to control group. Based on these finding studies, it is suggested that HEE can be developed as co-chemotherapeutic agent of breast cancer through inhibition of HER2 expression.
