Expression of miRNA-21 AND mRNA PTEN (Phosphatase and tensin Homologous) PLASMA IN BREAST CANCER PATIENTS BASED ON STATUS AND MOLECULAR clinicopathologic IN Hospital Dr. Sardjito
ABSTRACT: Breast cancer is a complex disease that is characterized by heterogenity in genetic changes. MicroRNA are small RNA molecules 18-22 nucleotides, known to play an important role in the regulation of expression at the level of physiology and pathology. MicroRNA (miRNA) are found in the plasma and has a character that is ideal as a biomarker for cancer detection. microRNA (miRNA) dysregulation in cancer, miRNA expression patterns in human plasma showed tissue-specific cancer, and miRNA have a high stability in plasma. MicroRNA-21 (miR-21) is the only miRNA that has increased expression in all human cancer including breast cancer. PTEN (Phosphatase and Tensin Homolog) is a tumor supressor gene deletion or mutation in a variety of human cancer. PTEN expression decreased in the majority of solid tumors including breast cancer and PTEN mRNA are known to be directly targeted by miR-21. In this study, we want to know miR-21 and PTEN mRNA expression at an early stage and advanced stage as well as in term of clinicopathologic status in the plasma of breast cancer patients at the RSUP Dr. Sardjito. Expression of miR-21 and PTEN mRNA was analyzed by Real Time qPCR. Present study showed the expression level of miR-21 in the advanced stage 1,32-fold higher than the early stage while PTEN mRNA expression level decreased 1,33-fold at an advanced stage compared to the early stage. MicroRNA-21 expression in term of clinicopathologic status obtained results are the highest expression in the luminal B subtype and overexpression in ER+. PR+, Her2+, while the highest expression of PTEN mRNA foung in Her2 overexpression subtype and overexpression in hormone receptor Her2+. In this study that there are differences in level expression of miR-21 and PTEN mRNA at early stage and advanced stage as well as in term of clinicopathologic status
