Expression of miRNA-155 AND OF suppressor mRNAs cytokines signaling 1 (SOCS1) BLOOD PLASMA IN BREAST CANCER PATIENTS BASED ON STATUS AND MOLECULAR KLINIKOPATOLOGIS

ABSTRACT: Background : Breast cancer still be important global health problems. miR-155 was reported had important role in the development of breast cancer. The role of miR-155 is targeting the mRNAs that has an important role as tumor suppressor genes, one of which SOCS1 mRNA. The discovery of miRNA in body fluids become a new hope for the development of minimally invasive cancer biomarker However, until now there has been no report on the expression of miR-155 and SOCS1 mRNA in plasma breast cancer patients based on clinicopathological and molecular status, especially in Indonesian population. Objective: This research try to analyze expressions of miR-155 and SOCS1 mRNA on plasma breast patients based from clinicopathological and molecular status. Methods: This study was conducted using cross-sectional design, sample that has been collected from Dr. Sardjito Hospital. RNA was extracted from plasma using miRCURY RNA Isolation Kit-Biofluid. cDNA synthesised using cDNA Synthesis kit II, 8-64 rxns running by PCR thermal cycler (Biorad c1000) and Real-time qPCR for detection of miR-155 expression using ExiLent SYBR Green master mix (Exiqon) by Biorad CFX 96. mRNA SOCS1 was analyzed by One-Step qRT-PCR using KAPA SYBR kit. Both expression was analyzed by Livaks method and independence T-test. Results: Expression of miR-155 at late stage was 2,426 higher than the early stages (p=0,047), while the mRNA expression of SOCS1 at late stage was 1, 294 lower than early stage (p=0,170). Expression of miR-155 at ER, PR, & HER2 status indicates different values, while the mRNA expression of SOCS1 was not different. miR-155 expression was higher in triple negative, while SOCS1 higher in luminal A. But, the difference are not statistically significant (p value > 0,05). Conclutions: Thus, this study proved that miR-155 and SOCS1 mRNA can be detected in blood plasma and both maybe could be prognostic biomarker in minimal invasive method.
