In Vitro Cytotoxicity test n-hexane extract, dichloromethane, AND ETHANOL SENGKUBAK (Pycnarrhena cauliflora (Miers.) Diels) CELL LINE IN CULTURE AND CANCER T47D selectivity OF VERO CELLS
ABSTRACT: Background : Breast cancer is one of the most common cancers and mostly affects women at various ages. Nowadays, a lot of research studied about herbs that can be used as an anticancer drug, including local plants from Kalimantan, Sengkubak (Pycnarrhena cauliflora (Miers.) Diels) which contain bisbenzylisoquinoline alkaloids that can induce apoptosis in cell line HeLa, but the effects on cell line T47D is unknown. Objective : Determine the cytotoxicity and selectivity of Sengkubak extracts in cell line T47D and Vero. Method : The method is post-test only with non-equivalent control group design. Each T47D and Vero cells were incubated in three kinds of Sengkubak extract (Pycnarrhena cauliflora (Miers.) Diels) in n-hexane, dichloromethane, and ethanol, in various concentrations. Cytotoxicity assay was performed with MTT assay. Results obtained in percentage of inhibition of cell growth, and then was analyzed statistically to determine IC50 value and selectivity index against Vero. Result : IC50 value of n-hexane, dichloromethane, and ethanol are 244,79 µg/mL, 49,63 µg/mL, and >1000 µg/mL respectively with selectivity index value against Vero are 0,66; 10,66; and undetermined respectively. Conclusion : Dichloromethane extract (IC50 49,63 µg/mL) has potency to be developed as cytotoxic agent and shows high selectivity index (IS) value, while n-hexane extract (IC50 244,79 µg/mL) and ethanol extract (IC50 >1000 µg/mL) has no potency to be cytotoxic agent.
