EXTRACT cytotoxic activity ethanolic wooden cup (Caesalpinia sappan L.) AGAINST BREAST CANCER CELL-TARGETED MOLECULAR RECEPTORS IN HER-2
ABSTRACT: Human Epidermal Receptor-2 (HER-2) has an important role in the initiation of breast cancer cells proliferation. Inhibition of HER-2 expression is a promising strategy in the breast cancer treatment. One of the potential natural ingredient is secang (Caesalpinia sappan L.) heartwood which contain flavonoids brazilin and brazilein. These compounds have been known to have cytotoxic activity against some breast cancer cells. The purpose of this study is to determine the cytotoxic activity of EES (ethanolic extract of the secang�¢ï¿½ï¿½s heartwood) on breast cancer cells which is targeted on HER-2 expression. EES was obtained by maceration assay using 70% ethanol. The active compounds were analysed using TLC. MTT assay was performed to determine the cytotoxic activity of EES with IC50 value as the parameter. The expression of HER-2 was observed by immunofluorescence assay. TLC results showed that EES contained brazilein with hRf value of 85. Single treatment of EES performed cytotoxic activity with IC50 value of 25 ���µg/mL on MCF-7/HER-2 breast cancer cells. With 12.5 ���µg/mL of EES (���½ IC50), it inhibited HER-2 expression on MCF-7/HER-2 breast cancer cells compared to control. Overall, EES has cytotoxic activity on breast cancer cells through HER-2 expression inhibition. Key word : Cytotoxic activity, Caesalpinia sappan L., breast cancer cells, HER-2 receptor
