TEST cytotoxic TAK FRACTION OF SOLUBILITY n-hexane LEAF EXTRACT ETHANOL ficus septica (Ficus septica Burm. F.) AND ITS ESSENTIAL alkaloid ON BREAST CANCER CELLS

ABSTRACT: Ethanolic extract of Ficus septica leaf showed potential cytotoxic effects on MCF-7 cells and enhanced the cytotoxic effects of doxorubicin. The insoluble fraction of n-hexane from ethanolic leaf extract to possess cytotoxic activity against breast cancer cells T47D. Ficus septica can be developed as breast anticancer agents. This study aims to examine the cytotoxic activity from insoluble fraction of n-hexane and the major alkaloid compounds in breast cells. Extraction process used 70%ethanol and was partitioned using n-hexane. The n-hexane insoluble fraction was reacted with the acid-base and partitioned with ethyl acetate. Separation and purification of major alkaloid compound from ethyl acetate fraction was performedby preparative TLC with mobile phase of 2-propanol:acetic acid:water (5:1:1) and silica PF254 as stationary phase. The purity of this compound were identified by TLC and HPLC-PDA and identified by LC-MS, IR and NMR. The next step was to test the cytotoxic activity of n-hexane insoluble fraction and major alkaloid compound againt breast cells and vero cells. The major alkaloid compound from n-hexane insoluble fraction with relative purity94.38% (TLC and HPLC). This compound was an alkaloid with m/z 360.5 contained of aromaticand methoxy subtituen with an odd number of nitrogen atoms present in the molecule. HIF had low cytotoxic effects and not selectiveonMCF-7 andT47DwithIC50valuesof each142,94 microgram/mL (SI 2,18) and435,89 microgram/mL (SI 0,69). The compound of MA-HIF exhibited cytotoxic effect and not selective with IC50 values of 68,08 microgram/mL (SI 1,54) and 49,32 microgram/mL (SI 2,12).
