ABSTRACT: The number of previous studies regarding the use of natural materials as a source of new medicines and the use of compounds from natural materials as superior chemotherapeutic agents is the basis of this study. Several classes of alkaloids contained in the plant have cytotoxic effects against cancer cells. The aim of the study was to determine the alkaloid profile of ethyl acetate fraction of ethanol -soluble component (FEA-KLE) of ethanolic extract of leaves of awar-awar (Ficus septica Burm. f.) and determine the potency of FEA- KLE as a cytotoxic agent against breast cancer cells T47D. In this study, FEA - KLE ethanolic extract of awar-awar leaves (Ficus septica Burm. f..) were obtained from several stages: maceration using ethanol 70 % v/v; filtration using aluminum oxide (Al2O3); residue extraction using ethanol 96 % v/v (ethanol soluble fraction/ FLE ); adding FLE with HCl 0.1 N, Mayer reagent, and 0,1 N NaOH and the partition using ethyl acetate (FEA - KLE). Alkaloid's profile substance obtained by qualitative analysis using thin-layer chromatography stationary phase such as silica gel GF254 plates and a mobile phase of n-butanol: glacial acetic acid: distilled water (3:1:1 v/v/v), and then tested with several alkaloid's reagents were Dragendorff and Van Urk Salkowski spray. MTT test was conducted to determine the cytotoxic activity of the FEA-KLE T47D breast cancer cells using the number of concentrations; 3,12 ;6.25 ;12.50 ;25 ; and 50 ug/ml . Analysis of alkaloid's profile using thin layer chromatography (TLC) with a stationary phase such as silica gel GF254 plates and a mobile phase of n- butanol: glacial acetic acid: distilled water ( 3:1:1 v /v/v) that showed the contains of alkaloids at Rx 0.74; 0.84 ;0.92 ;and one of the comparison yohimbine. Rx 0.84 had a positive reaction to Van Urk Salkowski reagent indicating indole alkaloids in FEA-KLE. The results of densitometry at 254 nm using a TLC scanner after elution with the stationary phase and mobile phase were similar, indicating alkaloid profiles in FEA-KLE amounted to 52.00 %, 22.69 %, and 25.31 %. Cytotoxic potential of FEA-KLE against T47D breast cancer cells obtained from the MTT assay results with FEA- KLE IC50 value of 2.73 ug/ml. Regarding of these results, we could conclude that FEA- KLE contains several classes of compounds including indole alkaloid, has high activity against breast cancer cell line T47D .
