GIVING EFFECT OF COMPOUNDS DERIVED isoflavones 1,2-EPOKSI- 3 [3- [3,4-DIMETOKSIFENIL] -4H-1-benzopyran-4-ON] propane on cell proliferation alveolar WHITE LUNG IN RATS (SPRAGUE Dawley) MODEL OF CANCER THAT INDUCIBLE DMBA

ABSTRACT: Backgrounds: Cancers the leading causes of death worldwide. Lung cancer ranks first contributor to cancer deaths. In cancer occurs increase of cell proliferation. Isoflavone is compounds that have potential as anticancer by inhibiting cell proliferation. In this research, isoflavone derivative compounds of clove oil (eugenol), 1,2-Epoksi-3[3-(3’4’-dimetoksifenil)-4H-1-benzopiran-4- on] propane (EPI) expected to decrease the proliferation of alveolar cells. Objectives: To know the effect of EPI compounds on alveolar cell proliferation of white rat (Sprague Dawley) cancer model which induced by DMBA with parameter IHC anti-PCNA. Methods: The subject used is 36 white rats (Sprague Dawley) female, 2 weeks old, and ± 40 gram in weight. Subject was divided into 6 groups randomly. Control group, fed only. Oil group, were given corn oil (solvent DMBA). DMBA group, were given DMBA 2x20mg/kgBB for 5 weeks, a rat model cancer. EPI 1 mg group, rat model cancers were given isoflavone compounds 1mg/kgBB for 15 weeks. EPI 2 mg group, rat model cancers were given isoflavone compounds 2mg/kgBB for 15 weeks. EPI 4 mg group, rat model cancers were given isoflavone compounds 4mg/kgBB for 15 weeks. After completion, the sample of lung was staining with IHC anti-PCNA. Results: DMBA increase the number of alveolar cells that express PCNA (p<0,05). Isoflavone 1 mg and isoflavone 2 mg does not decrease the number of alveolar cells that express PCNA (p<0,05). Isoflavone 4 mg decrease the number of alveolar cells that express PCNA (p>0,05). Conclusion: Isoflavone 4 mg able to decrease the proliferation of alveolar cells in the white rat (Sprague Dawley) cancer model which induced by DMBA.
