COMPOUNDS cytotoxicity assay ISR 7-MIDO SYNTHESIS OF BREAST CANCER CELLS IN CULTURE AND MCF-7 T47D

ABSTRACT: Background: Breast cancer is one of the most common cancer in the world who suffer in the women. Isoflavon is a compound of natural subtance that has anti-cancer activity. 7-mido ISR is an isoflavone derivate which is synthesized from eugenol, so it suspected has cytotoxic activity on cancer cell. Objective : This research done to know the effect cytotoxicity of 7-mido ISR which is synthesized from eugenol with count IC50 on breast cancer cell T47D and MCF-7. Method: This research was quasi-experimental study using cell line culture of breast cancer T47D and MCF-7. The experimental group was 7-mido ISR while doxorubicin was control group. Cytotoxic test used MTT assay method. Cytotoxic test done in the cancer cell which have been incubate for 24 hours use the experimental compound in various concentration 6,25; 12,5; 25; 50; 100 Ã���Ã�¯Ã�¿Ã�½Ã��Ã�¯Ã��Ã�¿Ã��Ã�½Ã���Ã�¯Ã�¿Ã�½Ã���Ã��Ã�µg/mL in each of the well in triplicate. The result has been tested using ELISA reader for reading the absorbance. Absorbance of each well was analyzed to calculate average percentage of cell growth inhibition. Then, the IC50 was calculated by probit regression analysis. Result: This research shows 7-mido ISR compound has cytotoxicity effect on the breast cancer cell line T47D with value of IC50 was 28,002 Ã���Ã�¯Ã�¿Ã�½Ã��Ã�¯Ã��Ã�¿Ã��Ã�½Ã���Ã�¯Ã�¿Ã�½Ã���Ã��Ã�¼g/ml and 13,190 Ã���Ã�¯Ã�¿Ã�½Ã��Ã�¯Ã��Ã�¿Ã��Ã�½Ã���Ã�¯Ã�¿Ã�½Ã���Ã��Ã�¼g/ml in MCF-7. Conclusion: 7-mido ISR compound has cytoxicity effect on breast cancer cell line T47D and MCF-7. Keyword: 7-mido ISR, isoflavon, IC50, T47D, MCF-7, Cytotoxicity.
