TEST COMPOUNDS cytotoxic activity of 7-O-propyl 3 ', 4'- DIMETOKSIISOFLAVON SYNTHESIS OF CELL CULTURE IN BREAST CANCER MCF-7 AND T47D
ABSTRACT: Background: Cancer is an illness with high incidence and high morbidity. Isoflavon is a compound of natural substance that can prevent many kind of chronic illness, especially cancer. A compound of 7-O-Propil 3’, 4’ Dimethoxyisoflavon is a new compound which is derived from isoflavon, but the cytotoxic activity of this compound on cancer cell is not known. Objective: This experiment was intended to know the cytotoxic activity of 7-O-Propil 3’, 4’ Dimethoxyisoflavon which is isoflavone derivate compound on cell line culture of breast cancer MCF-7 and T47D. Method: This research was a quasi-experimental study using cell line culture of breast cancer MCF-7 and T47D. The experimental group was 7-propa ISR while doxorubicin was the control group. Cytotoxicity test used MTT assay method. Experimental compound in various concentration 6,25; 12,5; 25; 50; dan 100 µg/mL was put into 96 wells that contain duplicative cell line and was incubated for 24 hours. The result of this research was read by ELISA reader. Absorbance of each well was analyzed to calculate the average percentage of cell growth inhibition. The IC50 value which is concentration level that inhibits 50% of cell growth was calculated by probit analysis. Result: The IC50 value of 7-O-Propil-3’, 4’- Dimetoksiisoflavon on cell line MCF-7 was not determined, however it was 83,263 µg/mL on cell line T47D. Conclusion: The 7-O-Propil-3’, 4’-Dimetoksiisoflavon compound has less cytotoxic activity on cell line culture of breast cancer MCF-7 and T47D than doxorubicin compound.
