Cytotoxic effect, proliferation, and APOPTOSIS FRACTION alkaloids ficus septica LEAVES (Ficus septica Burm.f.) AGAINST BREAST CANCER CELLS T47D

ABSTRACT: Ficus septica Burm.f. is a plant that potential to be developed as an anticancer agent. Alkaloid as the main compound exhibited cytotoxic activity on human cancer cell lines. Ethanolic extract from Ficus septica Burm. f. leaves exhibited cytotoxic activity against breast cancer cells T47D. This study aims to investigated the cytotoxic effect of alkaloids fraction from ethanolic extract on T47D cells as well as to identified the molecular mechanism. Ethyl acetate fraction (FEA) was obtained from the Ficus septica leaves ethanolic extract that fractionated gradually with Al2O3 and acid base reaction. FEA was separated by preparative TLC with mobile phase propanol: glacial acetic acid: water (5: 1: 1) and the stationary phase silica gel PF254. The separation obtained five sub fraction (SF) I-V. Alkaloid compound was analized using TLC with mobile phase propanol: glacial acetic acid: water (5: 1: 1) and the stationary phase silica gel 60 F254. The alkaloid from FEA and SF III were indicated as a red spot after sprayed with reagent Dragendorff. Cell viability was evaluated on T47D cells lines by using the MTT colorimetric assay. Alkaloid compound indicated in the FEA as well as SF I-V. The cytotoxic effect of FEA showed IC50 value 10.27 μg / mL, while SF I-V were found at 76.65; 44.78; 28.06; 37.20; 55.13 μg / ml, respectively (ANOVA-Tukey HSD Analysis) and doxorubicin as positive control at 0.076 μg/ml. These results exhibited that SF III (Alkaloid fraction) was the most active sub-fractions. Proliferation assay with incubation time extension for 24, 48, and 72 hours showed a decrease in cell viability up to 27.72% and induced cell death through apoptosis (16.7%) after treatment with SF III. Alkaloids fraction from Ficus septica leaf exhibited anti-cancer activity on breast cancer T47D cell lines.
