EFFECT OF COMPOUNDS DERIVED isoflavones 1,2-EPOXY-3- [3- (3,4-DIMETOKSIFENIL) -4H-1-benzopyran-4-ON] propane caspase-3 EXPRESSION OF THE BREAST CANCER MODEL RATS (SPRAQUE Dawley) THE INDUCIBLE DMBA

ABSTRACT: Backgrounds: Breast cancer is the most common cancer in women worldwide. Recently, several chemopreventive agents against breast cancer have been developed from plants, such as isoflavones. One of the chemopreventive mechanisms of isoflavones is apoptosis-induced. Apoptosis as a defense mechanism against cancer can be assessed from the expression of caspase-3 which is the main executor in the process of apoptosis. An isoflavone derivative compound 1,2-epoxy-3[3-(3,4-dimethoxyphenyl)-4H-1-benzopyran-4-on]propane synthesized from clove oil (eugenol) may have chemopreventive activity similar to isoflavones. Purpose: To know the effect of isoflavone derivative compound 1,2-epoxy-3[3-(3,4-dimethoxyphenyl)-4H-1-benzopyran-4-on]propane on the expression of caspase-3 in DMBA-induced Spraque Dawley rats breast cancer models. Methods: The subjects used in this study were 35 female Spraque Dawley rats, aged 2 weeks, with a weight of Ã�± 40 grams, divided into 7 groups randomly. The control group was fed only. DMBA group which is a rat model of cancer was fed and DMBA was also given to it. Corn oil group was given corn oil used as a solvent of DMBA. EPI 1 group was given DMBA and 1 mg/kgBW of EPI. EPI 2 group was given DMBA and 2 mg/kgBW of EPI. EPI 4 group was given DMBA and 4 mg/kgBW of EPI. DMBA was given 2x20 mg/kg per week from 6th to 10th week and EPI was administered from 2nd to 15th week. At the end of the 16th week, all rats were decapitated and the nodules were taken for histological preparations made with paraffin method and anti-caspase-3 IHC staining. Results: The expression of caspase-3 of EPI 1, EPI 2, and EPI 4 groups are significantly lower (P < 0.05) than DMBA group. DMBA group has the highest expression of caspase-3. There are no significant differences between the control group with EPI 1, EPI 2, and EPI 4 groups (P > 0.05). Conclusion: Isoflavone derivative compound EPI with a dose of 1 mg/kgBW, 2 mg/kgBW, or 4 mg/kgBW decrease the expression of caspase-3 in DMBA-induced Spraque Dawley rats breast cancer models.
